Initiation

Before initiating a CRISPR/Cas9 mouse project please:

  • Consult with the TGE Core regarding the preparation or purchase of these components and the targeting strategy under consideration.
  • Receive approval of the Institutional Animal Care and Use Committee (IACUC) and the Institutional Biosafety Committee (IBC) for your animal use and recombinant DNA/sgRNA protocols. Then submit your orders through Infinity.

Services

Generation of Knock-Out (KO) Allele by CRISPR-Mediated Genome Editing: $7,500

 

Service Description

  • Bioinformatics analysis of genomic locus to be targeted, as well as design, procurement, and in vitro validation of sgRNA function;
  • SgRNA/Cas9 delivery either by microinjection (MI) or electroporation (EP);
  • PCR-based genotyping of pups born to identify potential founder mice;
  • Breeding founder mice with C57BL/6J mice to heterozygosity and to determine germline transmission (GLT) of KO allele;
  • Delivery of mice (at least two heterozygous mice) carrying the desired KO allele;

Epitope (1X) Knock-In (KI) Allele Production by CRISPR/Cas9.: $8,900

 

Service Description

  • Bioinformatics analysis of genomic locus to be targeted for KI, as well as design and procurement of sgRNA and ssODN or dsODN donor DNA, in vitro validation of sgRNA function;
  • SgRNA/Cas9 and donor DNA delivery either by MI or EP;
  • PCR-based genotyping of pups born to identify potential founder mice;
  • Breeding founder mice with C57BL/6J mice to heterozygosity and to determine GLT of KI allele;

Point Mutation Production (Single Nucleotide or Amino Acid Codon Change) by CRISPR/Cas9.: $8,900 

Service Description

  • Bioinformatics analysis of genomic locus to be targeted, as well as design and procurement of sgRNA and ssODN or dsODN donor DNA, and in vitro validation of sgRNA function;
  • SgRNA/Cas9/donor DNA delivery either by MI or EP;
  • PCR-based genotyping of pups born to identify potential founder mice;
  • Breeding founder mice with C57BL/6J mice to heterozygosity and to determine GLT of desired mutated allele;
  • Delivery of mice (at least two heterozygous mice) carrying the desired mutated allele;
    Note: this service is limited to three MI/EP sessions with no guarantee of success if the mutation results in embryonic/postnatal lethality.

Gene Deletion (Up to 300 kb in size) by CRISPR/Cas9.: $8,500

 

Service Description

  • Bioinformatics analysis of genomic locus to be targeted for deletion as well as design, procurement, of a pair of sgRNAs, and in vitro validation of sgRNAs;
  • Delivery of pair of sgRNA/Cas9 either by MI or EP;
  • PCR-based genotyping of pups born to identify potential founder mice;
  • Breeding founder mice with C57BL/6J mice to heterozygosity and to determine GLT of deletion;
  • Delivery of mice (at least two heterozygous mice) carrying the desired genomic deletion;
    Note: this service is limited to three MI/EP sessions with no guarantee of success if the deletion results in embryonic/postnatal lethality.

Conditional Knockout (cKO) Allele Production by CRISPR/Cas9.: $13,500

 

Service Description

  • Bioinformatics analysis of genomic locus for placement of loxP sites, as well as design and procurement of a pair of sgRNAs and loxP ssODN or dsODN donor DNAs; in vitro validation of sgRNAs;
  • Delivery of pair of sgRNA/Cas9 and loxP donor DNAs either by MI or EP;
  • PCR-based genotyping of pups born to identify potential founder mice;
  • Breeding founder mice with C57BL/6J mice to heterozygosity and to determine GLT of cKO allele;
  • Delivery of mice (at least two heterozygous mice) carrying the desired cKO allele.

Knock-in (Less than Two kb) Mouse Production by CRISPR/Cas9.: About $14,500 to $20,000

 

Service Description

  • Bioinformatics analysis of genomic locus to be targeted for KI, as well as design and procurement of sgRNA and dsODN donor DNA, in vitro validation of sgRNA function;
  • SgRNA/Cas9, and donor DNA delivery either by MI or EP;
  • PCR-based genotyping of pups born to identify potential founder mice;
  • Breeding founder mice with C57BL/6J mice to heterozygosity and to determine GLT of KI allele;
  • Delivery of mice (at least two heterozygous mice) carrying the desired KI allele.

Conventional Transgenic Mouse Production (0.5 to 18 kb): $7,500 to $20,000

The price for this service greatly depends on the size of the transgene (tg) construct, and whether the PI provides the Core with a plasmid containing the tg or whether tg design and molecular cloning is the Core’s responsibility. Given the great variety in the type of tg experiments, we encourage the PI to consult with us to find the most sensible solution. Aside from this, the service will include:

  • Tg construct purification to obtain MI-ready DNA;
  • MI of tg construct;
  • PCR-based genotyping of pups born to identify potential founder mice;
  • Breeding founder mice with C57BL/6J mice to heterozygosity and to determine GLT of tg;
  • Delivery of mice (at least two heterozygous mice) carrying the tg.

DsDNA Synthesis and Purification for Microinjection: $1,000

DNA template will be synthesized and cloned into plasmid. Mid- or large preparation of plasmid DNA will be performed, and dsDNA fragment will be isolated and purified for microinjection. For DNA size over one kb, additional charge will apply at $0.5/bp over one kb.

PCR and Sequence Confirmation of CRISPR/transgenic Founders: About $500 to $1,000

The core will isolate tail DNA samples from founder pups, design and perform PCR genotyping, isolate PCR DNA fragments for sequencing, and identify the positive founders based on PCR and sequence results. This package price includes all reagents and two to three tech days and applies to one single microinjection session of one single mutation event.
